The main characters of the show, they are a division of a worldwide actually galactic organization known as the Kids Next Door. They are considered to be one of the best teams in the KND.

The leader of Sector V, an overworking agent who loves every minute of his job. He is widely considered one of the best of the best. However, his paranoia and stubbornness have gotten in the way of doing his job effectively at times. He is voiced by Benjamin Diskin.

The team's technology expert who loves cracking bad jokes. As Sector V's technology officer, he designs many of the 2×4 technology the team uses In his off time he enjoys building and flying airplanes and solving mysteries at his school. Voiced by Benjamin Diskin.

- Loving a Shadow: His crush on Cree is pretty superficial, given he focuses on her beautiful appearance rather than her unpleasant personality. It's also implied his crush was based on his own implicit feelings on his teammate and Cree's sister, Numbuh Five.
- Meaningful Name: Hoagie P. Gilligan Jr., appropriately enough, a twofer reference. First, there's "Gilligan," a reference to the title character of Gilligan's Island, who was also second-in-command on the S.S. Minnow. Then there's the "Jr.", often used at the end of a boy's name to signify that he's the second male in the family to have inherited his father's name (a seemingly throwaway detail that actually foreshadows the importance of his Disappeared Dad, Hoagie P. Gilligan, Sr.)
- Must Make Her Laugh: Many of his awful puns are done to make Numbuh Five laugh (she's instead very annoyed). This is the main hint of his feelings for her.
- Nice Guy: He does have his flaws, but he's still a very friendly and cheerful guy, and the only one who lacks the Anti-Hero attributes the others have.

The team's medic, and the most feminine of the kids, a happy-go-lucky girl who loves Rainbow Monkeys. In battle, she specializes in diversionary tactics. She seems to have a lot of admirers, but her heart obviously belongs to a certain Aussie. Voiced by Lauren Tom.

The team's combat specialist. He's incredibly lacking in book smarts, but his combat prowess more than makes up for it. In his off time, he competes in adult disapproved sports. Voiced by Dee Bradley Baker.

The team's second-in-command, she's the one that keeps a level head under all circumstances, even when everyone else loses it. In her spare time, she's a candy hunter, which brings her in conflict with her rival Heinrich and candy pirate Stickybeard. Voiced by Cree Summer.

- Agent Scully: Is usually skeptical toward Nigel's conspiracy theories and will often be wrong.
- Berserk Button: Her calm demeanor goes out of the window whenever Numbuh Two makes one of his awful puns, prompting her to hit him with her hat in response. Though it's implied part of it is a front to hide her feelings towards him, considering she makes bad puns herself whenever he's not present.
- Betty and Veronica: The Betty to her sister's Veronica for Hoagie's Archie. She wins.

After getting critically injured by Cree, Numbuh Two upgrades him into half-skunk, half-robot R.O.B.O.-B.R.A.D.L.E.Y. in "Operation: H.O.S.P.I.T.A.L.". He is voiced by Dee Bradley Baker.

The former Head of Decommissioning and current Global Tactics Officer. She's a shrill girl with a particular intolerance for the male sex. But due to her behavior, she has next to no friends. Voiced by Jennifer Hale.

- Also don't ever call her "Toots", as Numbuh 2 learned the hard way in "Operation: P.I.N.K.-E.Y.E."

- Well, meaningful Numbuh, when you look at her rank: to be "eighty-sixed" is American slang for being thrown out, eliminated, or rejected (it's also used in restaurant lingo to mean that a menu item has run out and is no longer available)
- Her first name is also meaningful, at least in the UK and Ireland where she is from. In the UK, "fanny" is a vulgar slang term for the female genitalia, another slang term for that part of the the body is "cunt" . "Cunt," in both UK and US slang, is also used to mean someone who is unpleasant (it's stronger than "bitch"), so combining that with Fanny's personality, and you have a Meaningful Name that somehow got past the censors -- at least the ones in the UK (in American English, "fanny" is one of many slang terms for the human buttocks and isn't considered obscene at all. Funny and a tad improper—yes, but not obscene).
- If you're willing to think deeper than her name could be meaningful in the US too. Another slang term for your butt is "ass", and that's even more commonly used to refer to an unpleasant person (similar to "bitch", once again).
- The Medic: A flashback to Numbuh Five's second Mission shows 86 wearing hat with a cross and checking on the injured. How she went from a medic to head of decommissioning is not explained.
- Never My Fault: Her introduction episode in which she never takes responsibility for her own blunders which wound up hampering Sector V's efforts for the mission. Thank karma kicked in when the spy she thought she she capture was actually an undercover agent (Number 362 in fact up) whose mission was ruined by 86 and get harshly chewed out for it (for added irony through out most of the episode she was putting down the boys in the group the most for any minor screw ups. End of the episode saw a girl doing this to her.)
- Noodle Incident: According to a canon comic, whatever made her hate boys happened after she became an operative.
- Slap Slap Kiss: With Numbuh Ninteenth Century.
- Straw Feminist: Later became less of this and more flat-out unfriendly to everyone.
- Too Dumb to Live: She's really dedicated to her job so much so that it blinds her to the most obvious points of logic such as in Operation E.N.D when she sent to capture and decommission Sector V based on the fact the computer says their turning 13. Numbuh One points out they're in the same age group, she and he even went to kindergardern together. But nope if the databanks says their teens it must be true! No point in questioning it.

Originally the top spy in the KND, she is now its current Soopreme Leader. Due to her busy and demanding job, she's often stressed, but at heart she stands stubbornly for the rights of kids everywhere, and if the situation arises she can be fun-loving too. Has grown to form a close friendship with Nigel Uno. Voiced by Rachael MacFarlane.

The mysterious head villain, Father is a man who wears a suit of shadows and is skilled in very dark powers. He schemes to enslave all children and turn them into his twisted version of "delightful". But despite his dark and malicious appearance, sometimes all he wants to do is curl up in a bathrobe with some Rocky Road. He is voiced by Maurice LaMarche.

As shown in the movie, when given the chance to fight against Grandfather alongside Numbuh Zero, B. Uno was too scared to do so and exits, not once but twice.
- Moral Myopia: Has own experiences of child abuse from his father, yet is perfectly fine with bullying kids during his grown up years.

Father's eerie children, supposedly an adult's view of the "perfect" kids. They are educated, well-dressed and polite, but are somewhat uncivilized in their burning hatred for the Kids Next Door. They always appear clustered together, move as one and speak in perfect unison. They are voiced by Benjamin Diskin, Dee Bradley Baker, and Cree Summer.

He then served as a high-ranking member of the Teen Ninjas, and lost all the respect he had to his name. Generally cocky and approving of himself, both before and after his betrayal. Voiced by Jason Harris.
- Becoming the Mask: Sort of. He is actually faking treachery and is a secret teen operative for the KND... however, after many defeats, his hatred for Numbuh One becomes real and he actually tries to kill him in his final appearance in the show.

What an Idiot!.
- Friendly Enemy: He was friends with Numbuh Four for awhile ("Operation M.O.V.I.E."), and joined the announcers in the finale. Though, in the CM/HB example he referred to Numbuh Four (actually Nightbrace) as his arch-nemesis. Gets kinda weird when it's revealed in a canon comic that he's Numbuh Four's uncle.

- Brought Down to Normal: Got turned into a chocolate monster that can turn anything he touches into chocolate. After Numbuh Five defeats him, the KND turns him back to normal.

- Gender Bender: In sacrificing his beauty, he switched genders. He's actually a girl.
- Greed: Obsessed with getting candy and never sharing it, which usually leads to many of his downfalls.

- Homage: To Indiana Jones Nazi villains.

- My Name Is Not Durwood: Numbuh Five calls him "Heinie".
- Never My Fault: He does this a lot, such as in his first appearance where he blames Abby for tainting the candy with the taste of asparagus when she warned him before hand that being greedy will put that curse into effect. It's also his grudge for the Guatemala Incident as after he sacrificed his beauty due to yet another curse. He once more blamed Abby for not stopping him from eating his caramels and supposedly leaving him stuck in that state. Turns out she was trying to find a way to break the curse.
- One-Winged Angel: In his first appearance after obtaining a jewel, he transforms into a monster with power of candy. Numbuh Five defeats him though. After falling into a chocolate volcano, he returned as a powerful monster who can turn things into chocolate by touching it. He was cured by the KND though.
- Noodle Incident: Throughout the series he remains angry at Numbuh Five for "Guatemala". His last apperance reveals that he performed a ritual to get delicious caramel by sacrificing their best trait (in his case beauty) and blamed Numbuh Five for allowing it to happen.
- Roaring Rampage of Revenge: When he returns after falling into a chocolate volcano, having been turned into a chocolate monster.
- Serious Business: Like with Stickybeard, Candy is serious business.
Grandfather

Father and Monty Uno/Numbuh Zero's father and an Evil Overlord who once ruled the entire planet. He's by far the most dangerous enemy the KND have every faced and makes Father look like a saint compared to him. Acted as the Big Bad in The Movie. Voiced by Neil Ross.
- Abusive Parent: He's why Father is the way he is.
- Big Bad: Of The Movie.
- Card-Carrying Villain: The first thing he says after being reawakened is to thank Father for "reawakening the pure evil" laying dormant within him.
- Eldritch Abomination
- Evil Overlord: Once ruled the world with an iron fist before Numbuh Zero betrayed and defeated him.
- Eviler Than Thou: To literally every villain in the Rogues Gallery once they find out his plan to turn everyone that's ever been a kid into Senor Citizombies includes them. Father actually outright states that Grandfather is eviler than him before reviving him, calling him "The World's Most Ultimate Evil".
- Expy: He seems to be partially based on Aku from Samurai Jack—a black, shadowy monster who stands a couple stories tall in his natural state, can manipulate fire, and his very awakening pretty much spells doom for the whole world.
- Fisher King: When he rules the world, he uses his Reality Warper abilities to convert into an old-style Crapsack World complete with child labor, Industral Era-style factories making tapioca pudding for him to eat.
- Genre Savvy: Is smart enough to NOT underestimate Hope Springs Eternal and understands he hasn't really won until he's destroyed the Book of KND, which let Numbuh Zero defeat him the last time. And he also doesn't go out to find it until nearly the entire world is under his control and the Kids Next Door are all but destroyed.
- Humanoid Abomination:
- Large Ham: Is an even bigger ham than his son.
- Laser-Guided Amnesia: Literally. Had his memory erased by Numbuh Zero using some kind of laser, reducing him to a harmless old man until he's "recommissioned" by Father. This actually seems to be the only way to defeat him, as all other attempts failed completely.
- Morality Pet: Subverted. Monty Uno/Numbuh Zero seems to be this, as he does like him as his son and considers him the favorite... but then tries to incinerate him when he turns on him without blinking an eye.
- Actually he liked Zero more for the fact he was willing to stand up to him and give Grandfather something of a challenge.
- Nigh Invulnerability: His reaction to getting crushed under the entire Kids Next Door Moonbase is to dust himself off and ask if they really thought that would work.
- No Ontological Inertia: His defeat restores the world to normal.
- Physical God: He's the single strongest villain in the series and is capable of surviving a direct hit from a Colony Drop without even a scratch. He can also 'age' the entire world at will.
- Playing with Fire: Father gets it from him.
- Rapid Aging: His primary ability is to age everything he touches, though not only does he age them rapidly, he turns them into his 'Senor Citizombie' minions.
- Reality Warper: He's capable of changing the world as he sees fit, normally to a more "old time" look.
- Super-Powered Evil Side: His human form is a harmless old man so long as his memories aren't restored.
- Trademark Favorite Food: Tapioca pudding, so much so that was one of the reasons he wanted to rule the earth.

Numbuh One's demanding girlfriend. While she's quite clingy, she does genuinely care for Nigel, but his work for the Kids Next Door strains their relationship. Voiced by Grey DeLisle.

- Betty and Veronica: Technically, she's the Betty to Rachel's Veronica, but she has more of a Veronica personality.
- Characterization Marches On: In her first appearance, she brainwashed Nigel into being her obediant boyfriend and even tried to make him destroy to his friends. The KND fight back and Nigel is unbrainwashed, but the episode ends with Lizzie getting the date she wanted anyway. Later episodes more or less completely ignored her origin story, and considerably softened her character.

- Easily Forgiven: Apparently Nigel and the rest of Sector V had no hard feelings over the little matter of her having originally tried to Brainwash him. They just don't verbalize it much due to fearing her bad temper (well, at least Numbuh Two fears her temper). Numbuh Five is tolerant of her at best.
